Abstract
Mutation of the Arabidopsis thaliana tRNA(CCA)(Trp) anticodon or of th e A73 discriminator base greatly diminishes in vitro aminoacylation wi th tryptophan, indicating the importance of these nucleotides for reco gnition by the plant tryptophanyl-tRNA synthetase. Mutation of the tRN A(CCA)(Trp) anticodon to CUA so as to translate amber nonsense codons permits tRNA(CUA)(Trp) to be aminoacylated by A,thaliana lysyl-tRNA sy nthetase, Thus, translational suppression by tRNA(CUA)(Trp) observed i n plant cells includes significant incorporation of lysine into protei n.
