Affected sib-pair methods are commonly used to detect linkage to dicho
tomous traits. This review discusses the principles underlying affecte
d sib-pair tests for linkage and the merits of sib-pair methods relati
ve to traditional fod score methods. Extensions of the methods to incl
ude other typed unaffected relatives, extra affected siblings, and mul
tiple marker and disease loci are reviewed together with an investigat
ion of the most effective ways to perform genome scans.
