B. subtilis membrane proteins were separated by reverse phase chromato
graphy using a simple solvent system of acetonitrile/water. The isolat
ed proteins were further characterized by a 'MS-FIT' peptide mass fing
erprinting program. The experimental scheme for separation and identif
ication of membrane proteins could also be modified to detect protein-
protein interactions. The method has been proven useful for rapid isol
ation, purification, and characterization of hydrophobic membrane prot
eins.
