A new liquid piezoelectric immunoassay was developed to determine infe
ction rabbit serum(IRS) by adult worm antigen (AWA) of S, japonicum, w
hich is based on the agglutination of antibody and antigen-PEG complex
by an immunoreaction. By examining the surface appearances of the qua
rtz crystal with scanning electronic microscopy(SEM) technology and in
vestigating the effect of the density and viscousity of the solution,
it can be found that the absorption on the surfaces of the quartz crys
tal plays a dominant role in the frequency response in the immunoassay
. The influences including PEG concentration, complexing time of AWA-P
EG complex and detection temperature on frequency responses were inves
tigated in detail, respectively. The frequency shift was linear with t
he antibody concentration in the range of 3.6 similar to 42.0 mu g/mL.
Five rabbit serum specimens were analyzed with this method and the an
alytical results given by this approach show that the detection ratio
of positive serum reaches 100% and the antibody concentration can be q
uantitatively determined.
