Abstract
Transverse thin cell layers (tTCLs) (0.2-0.4 mm thick) excised from the pse udo-bulblets of L. longiflorum in vitro were placed on the Murashige and Sk oog medium supplemented with 3% sucrose and a concentration of N-(2-chloro- 4-pyridyl)-N'-phenylurea (forchlofenuron, CPPU) varying from 0.1 to 10 mu M . Buds were obtained from the tTCL pseudo-bulblet explants cultured in the presence of CPPU in the light but not in the dark. The number of buds forme d per tTCL was the highest with 1 or 3 mu M CPPU (16.8 +/- 1.1 and 16.2 +/- 1.6 buds per tTCL, respectively). Testing a combination of sucrose (from 1 to 12 %) and CPPU (from 1 to 10 mu M) confirmed that the number of buds pr oduced per tTCL was the highest when 1 or 3 % sucrose was combined with 1 o r 3 mu M of CPPU. The regenerated shoots were transferred to soil after roo ting with 10 mu M indole-3-byturic acid and normal plants were produced. (( C) Academie des sciences / Elsevier, Paris.)
