The capability of the Klenow fragment deficient in 3'-->5' exonuclease acti
vity (KFexo(-)) to incorporate 2'-deoxyisoinosine (disoI) opposite canonica
l bases and 5-methyl-2-deoxyisocytidine (d(5Me)isoC) was examined. The trip
hosphate of disoI is incorporated opposite d(5Me)isoC designed as its pairi
ng partner. It is also incorporated opposite T and G according to a pairing
scheme involving its minor tautomeric form (the (OH)-H-2 tautomer).
