Salt-stress specific genes in Zygosaccharomyces rouxii detected by a differ
ential display method were analyzed, Total RNAs were extracted from Z. roux
ii cells cultured in YM broth containing 0% or 20% NaCl. cDNAs synthesized
from total RNAs were amplified by PCR, and the PCR products were separated
by polyacrylamide gel electrophoresis. Twenty-five genes specifically expre
ssed in Z, rouxii cells cultured in the absence or presence of 20% NaCl wer
e finally cloned into a TA vector and sequenced. Analysis of these genes re
vealed that 13 genes had no sequence similarity and 12 genes had similarity
to yeast genes present in the current GenBank/EMBL/DDBJ/PDB database searc
hed with the BLAST program. Among 12 genes similar to, yeast genes, 10 gene
s had similarity to Saccharomyces cerevisiae genes, one gene to a Candida a
lbicans gene, and one gene was the Z. rouxii plasma membrane H+-ATPase gene
