A single generic microbore liquid chromatography diode array/time-flight ma
ss spectrometry method has been developed that provides accurate mass on se
parated compounds cleaved from single beads and/or the decoding of orthogon
al tags pertaining to the compound in one run. The method is suitable for e
ncoded or non-encoded libraries. Accurate mass measurement within 5ppm can
be routinely achieved allowing differentiation of isobaric compounds in deg
enerate libraries. Accurate mass chromatograms can be used to resolve tag r
esponses from polyethylene glycol background interference. Accurate isotope
mass differences coupled with intensity ratios can be used for highly spec
ific isotopic target analysis for compounds of unknown molecular weight but
with a characteristic accurate isotope pattern. Accurate instrument calibr
ation is not necessary with this approach to target analysis. Copyright (C)
