Abstract
Menin is a protein product of a tumor suppressor gene MEN1, which is respon
sible for multiple endocrine neoplasia type 1, an autosomal dominant famili
al cancer syndrome. Rabbit antisera against human menin were generated by i
mmunization of synthetic peptides corresponding to human menin (206-230), (
471-488), and (576-594). These region-specific antisera thus produced were
found to be valuable for detection of immunoreactive menin by immunoblottin
g and immunocytochemistry in culture cells transfected with a menin-express
ing plasmid. Antibody binding was completely abolished by preincubation of
the antisera with respective antigens. The present study employing region-s
pecific antibodies confirmed the previous findings that menin is located pr
imarily in the nucleus. These antisera will be useful to investigate the sy
nthesis and metabolism of protein product of the normal and mutant MEN1 gen
es.
