The aim of this study was to establish a cost-effective and timesaving
method for the routine diagnosis of kappa-casein genotypes of breedin
g bulls. DNA of 65 animals was isolated from semen by alkali lysis. Th
e genotype was analysed by a single tube allele-specific (STAS) polyme
rase chain reaction (PCR). The results showed complete agreement with
those of an established method, which is more costly.
