Characterization of 1.8 kb Hind III fragment of Clostridium septicum genomic DNA

A genomic library of Clostridium septicum (IVRI strain 51) was generated by cloning Hind III fragments of genomic DNA of C. septicum into the plasmid pUC18. Thirty-three recombinant clones were screened and a recombinant plas mid (pSDS12) containing a 1.8 kb insert was selected for characterization b y restriction mapping. The map was confirmed by probing the restriction dig ests of the insert with 1.8 kb insert labeled with Digoxigenin-d-UTP. This insert has a unique Nsi 1 site. This Nsi 1 site could facilitate further su b-cloning as preliminary studies indicate that this insert may show utility as a gene probe in the diagnosis of C. septicum infections.
