The N6-adenine DNA methyltransferase M.TaqI binds duplex oligodeoxynucleoti
des containing 1,2-dideoxy-D-ribose at the target or 3' neighbouring positi
on with 200fold and 60fold higher affinity, respectively, compared to the n
atural substrate. These results are explained thermodynamically by a base f
lipping mechanism of M.TaqI.
