Abstract
The use of 1,2-dioxetane chemiluminescent substrates, including AMPPD(R), C SPD(R), CDP(R), and CDP-Star(R), with alkaline phosphatase enzyme labels pr ovides highly sensitive detection for numerous immunoassay and nucleic acid detection formats, These include membrane-based immunodetection of protein s, nucleic acid blot hybridization, immunoassays, and nucleic acid capture/ hybridization assays performed in a microwell plate, Enzyme cleavage of the 1,2-dioxetane substrate generates a metastable anion intermediate that dec omposes with the concomitant emission of light or transfer of energy to a f luorescent enhancer compound, Light emission exhibits glow kinetics, enabli ng the use of multiple imaging platforms for signal detection, including fi lm-based, luminometers, low-light sensitive camera, and phosphor screen ins trumentation systems.
