Abstract
To develop the genetic markers linked to the fertility restoring gene (Rf) found in continuous backcross progenies between Allium gaianihum Kar. et Ki r. (cytoplasm donor) and A. fistulosum L., isozymes and RAPDs were analyzed in the male fertile and male sterile plants in the backcross progenies (B- 3 similar to B-5). in isozyme analysis of six enzymes which showed interspe cific polymorphisms between A. galanthum and A. fistulosum, all the male fe rtile plants in the B-3, B-4, and B-5 generations had both alleles of A. ga lanthum and A. fistulosum at the phosphoglucoisomerase isozyme gene locus P gi - 1, whereas all the male sterile plants had only the allele of A. fistu losum. These results indicate that the Pgi - 1 of A. galanthum is linked to the Rf locus. In RAPD analysis using 27 random decamer primers which showe d interspecific polymorphisms between A. galanthum and A. fistulosum, all t he male fertile plants had an A. galanthum-specific band (OPE03(700)), wher eas none of the male sterile plants had this band. These results indicate t hat the OPE03(700) is linked to the Rf locus. These two genetic markers dev eloped in this study will be useful to identify the genotypes at the Rf loc us.
