Alternating films constructed upon successive deposition of redox polymer O
s(bpy)(2)ClPyCH(2)NHpoly (allylamine) and antibiotin IgG were developed for
amperometric immunosensor design. Cyclic voltammetric measurements were us
ed to verify charge transport between redox sites and the redox surface con
centration was estimated upon voltammetric peak integration. Biotin-antibio
tin complex formation was evaluated using horseradish peroxidase as an enzy
me label. Redox mediation between the modified electrode surface and the re
dox site in the enzyme was verified after substrate addition. Multilayer mo
dified gold electrodes with biologically active antibiotin IgG molecules we
re employed for the development of a competitive immunoassay with electroch
emical label detection. (C) 1999 Elsevier Science S.A. All rights reserved.
