The method was developed to attach a variable number of anthracene-containi
ng photoactive groups to protein molecules. Active reagents 9-anthryl diazo
methane and 9-anthrylmethyl isocyanate were allowed to react with carboxyl
and amino groups of proteins, respectively, in a heterogeneous reaction. Wi
th lysozyme and carboanhydrase B, a possibility was shown of studying the p
rotein dynamics and structure with luminescence spectroscopy by varying the
number of photoactive groups (on average, from 0.1 to 2.0) per protein mol
ecule.
