This study explores the feasibility of using triblock copolymer polyampholy
tes as a displacer to separate beta-lactoglobulin A and B isoforms by anion
exchange displacement chromatography. The displacers used herein are metha
crylic polyampholytes: A(8)M(12)B(12), A(16)M(12)B(12) and A(16)M(24)B(24),
where A is methacrylic acid, B is 2-(dimethylamino)ethyl methacrylate, and
M is methyl methacrylate. In addition, the steric factor, characteristic c
harges and binding equilibrium constant with the ion exchanger gel of the t
riblock copolymer are measured and discussed with the structure and solutio
n behavior of the polyampholytes. With the Steric-Mass Action (SMA) model o
f ion-exchange displacement chromatography, the elution and displacement li
ne can be constructed for the system. Moreover, the behavior of elution and
displacement line is qualitatively discussed and successfully proven.
