Tocopherols and tocotrienols are being increasingly recognized to have an i
mportant role in the prevention of atherosclerosis. It has been reported th
at they protect low-density Lipoprotein (LDL) and tissues from oxidative st
ress and that tocotrienols can reduce plasma cholesterol levels. Two isocra
tic high-performance liquid chromatography (HPLC) methods for simultaneous
analysis of tocopherols, tocotrienols, and cholesterol in muscle tissue wer
e developed. Method A involves basic saponification of the sample, but caus
es losses of the gamma- and delta-homologs of vitamin E. Method B does not.
involve saponification, thereby protecting the more sensitive homologs. Bo
th permit rapid analysis of multiple samples and neither requires specializ
ed equipment. These methods may provide techniques useful in simultaneous a
ssessment of oxidative stress status (OSS) and cholesterol levels. (C) 1999
