Ra. Farrer et al., Single-molecule detection with a two-photon fluorescence microscope with fast-scanning capabilities and polarization sensitivity, OPTICS LETT, 24(24), 1999, pp. 1832-1834
We describe a laser-scanning two-photon fluorescence microscope that is cap
able of observing single molecules with excellent temporal resolution and t
hree-dimensional spatial resolution. To demonstrate the capabilities of the
instrument we present. single-molecule fluorescence data obtained in sever
al different scanning modes. In addition, a polarization-sensitive detectio
n scheme can provide detailed three-dimensional information about the orien
tations of molecules in any of these scanning modes. (C) 1999 Optical Socie
