The effects of phenytoin and its major metabolite, 5-(4-hydroxyphenyl)-5-ph
enylhydantoin (HPPH), on bone resorption of neonatal mouse calvaria were ex
amined in vitro. Both phenytoin and HPPH induced significant bone resorptio
n as compared to the controls after 72 h in culture. This effect may be the
cause of phenytoin-induced bone loss in vivo.
