Abstract
We describe here the construction of a plasmid that combines positive selec tion with tetracycline resistance. The vector comprises a modified version of the gene;:encoding the cytosine-specific DNA methyltransferase MspI and a modified form of the pBR322 tetA(C) gene. The combination of these two ge nes facilitates the selection of recombinant plasmids in broth cultures, th ereby eliminating the need for bacterial plating. (C) 2000 Academic Press.
