Fluorescence correlation spectroscopy was used to characterise the conforma
tional equilibrium in "Calcium Green 1", a fluorescent indicator of calcium
. In the calcium bound form, a triplet bunching time of 6.7 mu s is seen, W
e show that, in the absence of calcium, 10% of the indicator is in a fluore
scent conformation that inter-converts to a non-fluorescent form in 10 ps.
This confirms the probing mechanism based on conformation controlled PET pr
oposed for Calcium Green 1.
