Radiosyntheses of [cyano-C-11]benzonitriles, [carboxyl-C-11]benzoic ac
ids, [carbonyl-C-11]benzamides and [tetrazoyl-C-11]5-biphenyI-1H-tetra
zoles are described. [Cyano-C-11]benzonitriles were prepared using the
Rosenmund-von Braun reaction in one step from [C-11]hydrogen cyanide
via [C-11]copper(I) cyanide in 2 min. [Carboxyl-C-11]benzoic acids and
[carbonyl-C-11]benzamides were prepared from [cyano-C-11]benzonitrile
-complexes using basic hydrogen peroxide for 3 min in N,N-dimethylform
amide at 180 and 80 degrees C, respectively. [Tetrazoyl-C-11]S-bipheny
]-1H-tetrazoles were obtained from [cyano-C-11]benzonitrile-complexes
in 7 min by cyclization with trimethyltin azide followed by acid hydro
lysis at 180 degrees C. These labelled compounds were obtained in a on
e-pot procedure with good average specific radioactivities [500 mCi/mu
mol (18.5 GBq/mu mol) at the end of synthesis] and in satisfactory av
erage radiochemical yields (24-69%) at 40 min from the end of radionuc
lide production. This new radioactive precursor, [C-11]copper(I) cyani
de, should give access to new C-11-labelled ligands for positron emiss
ion tomography. (C) 1997 Elsevier Science Ltd.
