Anther derived spontaneous doubled haploid (DH-R-1) plants were developed f
rom an intercultivar F-1 hybrid (Holland) of <blocky> type pepper cv. <Mazu
rka> (Capsicum annuum L. var. annuum, 2n = 24). Regenerants were grown in g
reenhouses for seed production to R-2-generation (DH-R-2) prior to flow cyt
ometric analysis which revealed DH-R-1 plants with haploid (n = 12); double
d haploid, DH, (2n = 24); tetraploid (2n = 48); and aneuploid genomes. Poll
en grains of anther cultures showed a high decrease in viability during the
first two weeks of incubation time. The DNA samples of the 47 DH-R-2 indiv
iduals were bulked in nine groups and subjected to PCR analysis for estimat
ing the level of homogeneity versus heterogeneity in comparison to the cont
rol of anther donor bulk of five plants of F-1 hybrid pepper cv. <Mazurka>
(DH-R-0). in total 26 primers were applied, 22 of them revealed scorable PC
R profiles with 54 scorable PCR bands. Of these, 19 (35.2%) bands generated
by 7 primers were polymorphic. All of the nine bulks of DH-R-2 plane were
discriminated at least by one primer.
