PCR-RFLP analysis was applied to classify nine Lilium species, L. japo
nicum, L. rubellum, L. nobilissimum, L. speciosum, L. auratum, L. aura
tum var. platyphyllum, L. henryi, L. concolor var. pulchellum and L. l
eichtlinii var. maximowiczii, and one interspecific hybrid, L. x formo
longi. When PCR products amplified in specific regions of ribulose-1,
5-bisphosphate carboxylase large subunit (rbcL) gene in chloroplast an
d nuclear ribosomal RNA (rRNA) gene were digested by 20 restriction en
zymes, some polymorphisms were observed. The examined plants were clas
sified into six groups based on the combined results in rbcL and rRNA
gene. The tested species in Archelirion and Sinomartagon were classifi
ed into different groups. Furthermore six species in Archelirion were
classified into three groups. Thus our study suggested that PCR-RFLP a
nalysis could be used for classification of lilies.
