Abstract
Direct shoot regeneration was induced from leaf explants of Arachis, grown in vitro. Shoot buds appeared at the cut surface of the vein within 8-15 da ys of culture and developed into shoots within 25-30 days. Among the differ ent concentrations of BAP used (1 to 6 mg/l), MS medium supplemented with 4 mg/l BAP along with 0.5 mg/l IAA gave high frequency of shoot bud initiati on (62.19%). Gradual increase in shoot induction (32 to 62%) was noticed wi th an increase in BAP (1 to 4 mg/l). Whereas at higher concentrations of BA P (5 to 6 mg/l), there was a decline in shoot initiation (57 to 50%). Regen erants with 1 to 3 cms in height were transferred to rooting media and 78% rooting was observed on MS media supplemented with 4 mg/l IAA + 0.5 mg/l KN . The rooted plants were transferred to soil and showed 100% survival rate.
