Abstract
We have studied the effect of chloridazone (in vitro 50 mu g/mg protein) on liver mitochondria by measuring oxygen consumption, and activity of ATPase activity, superoxide dismutase and glutathione reductase. Simultaneous mon itoring of mitochondrial respiration and membrane dynamics was performed by the method of synchronous fluorescence fingerprint. The inhibition of mito chondria respiration under condition of FAD-dependent substrate (succinate) occurred in both states after the addition of chloridazone, while the effe ct was different in the case of a NAD-dependent substrate (glutamate-malate ). The synchronous fluorescence fingerprint technique reveals changes in th e space arrangement of the mitochondrial membrane after the addition of chl oridazone, depending on the character of the substrate. The ATPase activity increased, that of superoxide dismutase slowly decreased and that of gluta thione reductase increased.
