Isoprostane E-2 (8-iso-PGE) and F-2 alpha (8-iso-PGF) sensitize nociceptors
and capsaicin-sensitive DRG neurons. In this study we investigated the cyc
looxygenase-dependence of isoprostane-induced C-nociceptor sensitization. S
ystemic pretreatment of rats with ketorolac (1 and 10 mg/kg) abolished 8-is
o-PGF sensitization and reduced the effects of 8-iso-PGE. Ibuprofen (30 mg/
kg) blocked all sensitizing effects. These data suggest that some algesic p
roperties of isoprostanes are mediated via prostanoid synthesis. (C) 2000 E
