We report the first simultaneous measurements of pH(i), [Ca2+](i) and tensi
on, upon alteration of pH(i), in isolated rat mesenteric arteries loaded wi
th both carboxy SNARF and indo-1. In these vessels (pre-contracted by 30 mM
KCl) alterations of pH(i), by addition and subsequent washout of weak base
s, produced complicated effects on tone. Although the changes in contractil
ity did not mirror the changes in pH(i) they were, at all times, accompanie
d by parallel changes in [Ca2+](i).
