Recombinant human acidic fibroblast growth factor (FGF-1) was radiolabeled
with Tc-99m by the HYNIC method. The Tc-99m-FGF-1 retained its representati
ve molecular mass, heparin affinity, cellular binding to both law (Kd = 9.5
nM) and high (Kd = 125 pM) affinity sites, and mitogenic activity. Gamma c
amera imaging after intravenous dosing in rats confirmed high liver and kid
ney binding. Heparin significantly decreased Tc-99m-FGF-1 liver uptake and
increased urinary excretion. These studies illustrate a new method for imag
ing FGF-1 targeting under various conditions. NUCL MED BIOL 27;4:407-414, 2
