Special modes of development such as unidimensional multiple development (U
MD), stepwise gradient development, and manual operation of automated multi
ple development (AMD, reversed gradient) have been used for the microprepar
ative separation of tertiary and quaternary alkaloid mixtures from Chelidon
ium majus L. roots. When the separations were compared with analytical-scal
e separations of identical mixtures it was found that the quaternary or ter
tiary alkaloids are well separated by use of chromatographic systems of the
type silica - multicomponent mobile phase and alumina (basic) - methanol dichloromethane mixture. The best results from zonal micropreparative sepa
ration of the alkaloid mixtures were obtained by use of unidimensional trip
le development and stepwise gradient elution.
