Electrophysiological and antiarrhythmic effects of a novel class III a
ntiarrhythmic drug nibentan were studied in 10 patients with paroxysma
l ventricular tachycardia. After intravenous injection of nibentan (0,
125 mg/kg) prolongation of QT and QT(c) intervals, significant increas
es of right atrial, right ventricular and His-Purkinje system's effect
ive refractory period were observed. Antiarrhythmic activity of nibent
an in relation to reproducible at endocardial pacing paroxysmal ventri
cular tachycardia was 80%. In 1 patient arrhythmogenic effect of the d
rug - polymorphic ventricular tachycardia (torsade de pointes) - was r
egistered.
