Specific features of analysis of the nuclear "trypsin-like" and "inhibitor-trypsin'' coleoptile complexes at the postembryonic phase of wheat development

"Inhibitor-trypsin'' and "trypsin-like" complexes of the nuclear matrix of
actively growing wheat coleoptiles and germinated wheat embryos were isolat
ed using affine chromatography on columns with immobilized trypsin and tryp
sin inhibitor, respectively. No such complexes were found after the termina
tion of the coleoptile growth or in the air-dry quiescent wheat embryos. El
ectron microscopy of the "trypsin-like" complexes has shown a rosette- and
bead-like structure with a dense granular layer which was absent in the "in
hibitor-trypsin'' complexes. Electrophoretic analysis demonstrated a range
of protein molecular masses from similar to 14 to similar to 67 kDa. DNA, R
NA. Carbohydrates were also found in the complexes. Immunoenzymatic analysi
s visualized endogenous auxin in the complexes of 24-hour germinated wheat
embryos.
