The regional brain kinetics of the two enantiomers of the NMDA channel
blocker ketamine radiolabelled with C-11 was studied in the Rhesus mo
nkey by means of positron emission tomography (PET). The uptake in bra
in areas which showed high radioactivities was blocked in a dose-depen
dent manner for both C-11-labelled enantiomers with simultaneous doses
of the respective unlabelled (S)- or (R)-ketamine, indicating specifi
c binding. The binding in the striatum and cortical areas of (S)-[N-me
thyl-C-11]ketamine was selective and displaceable by the (R)-enantiome
r and by MK-801.
