In the contest of the EUROFAN programme, me report the deletion and functio
nal analysis of six open reading frames (ORFs) on the right arm of chromoso
me XII of Saccharomyces cerevisiae. Using a PCR-based gene replacement stra
tegy, we have systematically deleted individual ORFs and subjected the hete
rozygous diploids and haploid knockout strains to basic genetic and phenoty
pic characterization. Two ORFs, YR127c and YLR129w, are essential for viabi
lity, whereas no growth phenotype could be detected following deletion of Y
LR124w, YLR125w, YLR126c or YLR128w. For each of the individual ORFs, a kan
MX4 replacement cassette and the corresponding cognate mild-type gene mere
