Abstract
We have previously characterized the first CD4-independent HIV-I X4 isolate m7NDK and showed a direct byding of this mutant gp120 with CXCR4. We prese nt evidence of down-regulation of CXCR4 membrane expression in CD4 positive cells chronically infected with the HIV-I m7NDK. This was shown by FAGS an alysis and the phenomenon was associated with a drastic reduction of steady state mRNA level. Kinetic analysis showed that CXCR4 is down-regulated in CEM cells within a maximum of six days post HIV-I m7NDK infection. Expressi on of the envelope protein might be responsible for this negative modulatio n of CXCR4.
