Triticum polonicum (2n = 28) and T. aestivum (2n = 42) were found to posses
s different types of interphase nuclei and prophase chromosomes when staine
d with orcein. T. polonicum showed the simple chromocenter or reticulate ty
pe of interphase nuclei and the interstitial type of prophase chromosomes,
whereas diffuse type of interphase nuclei and gradient type of prophase chr
omosomes were found in T. aestivum. The centromeric formulae of T. polonicu
m and T. aestivum were 18 m + 10 sm and 30 m + 12 sm, respectively. Six sat
ellites were observed in T. polonicum and five in T. aestivum when stained
with orcein. Only one satellite was found in both the species in CMA-staine
d materials. Two satellites appeared in DAPI-stained metaphase plates of T.
polonicum. These data indicate the stain specific nature of satellites in
these two species. Eleven CMA-positive bands were found in the nine chromos
omes of T. aestivum whereas no CMA-positive band appeared in T. polonicum.
DAPI band was not found in these species at metaphase. However, at prometap
hase eight DAPI-positive bands appeared in eight chromosomes of only T. pol
onicum. Therefore, The karyotype analysis with differential staining may ch
aracterize the chromosome complement of T. aestivum and T. polonicum.
