Comparative Tc-99m-MIBI, Tc-99m-tetrofosmin and Tc-99m-furifosmin uptake in human soft tissue sarcoma cell lines

Abstract
The uptake characteristics of technetium-99m hexakis-2-methoxyisobutylisoni trile (MIBI), Tc-99m-tetrofosmin and Tc-99m-furifosmin in human soft tissue sarcoma cell lines were investigated and compared. After 10-120 min of inc ubation at 37 degreesC, 32 degreesC and 22 degreesC with Tc-99m-MIBI, Tc-99 m-tetrofosmin and Tc-99m-furifosmin, the kinetics of cellular uptake of the se tracers in human soft tissue sarcoma cells SW 684 (fibrosarcoma), SW 872 (liposarcoma), SW 982 (synovial sarcoma) and SW 1353 (chondrosarcoma) was assessed. The uptake of Tc-99m-MIBI, Tc-99m-tetrofosmin and Tc-99m-furifosm in was temperature dependent. The kinetics of uptake of Tc-99m-MIBI and of Tc-99m-tetrofosmin was similar between fibrosarcoma and liposarcoma cells, as well as between synovial sarcoma and chondrosarcoma cells. Tc-99m-furifo smin showed similar uptake kinetics in all cell lines. The uptake of Tc-99m -furifosmin was, however, significantly higher in liposarcoma than in the o ther cells. The data indicate that the cellular uptake of Tc-99m-MIBI, Tc-9 9m-tetrofosmin and Tc-99m-furifosmin is dependent on cellular metabolic act ivity.
