The method of enrichment culture modified by the addition of sodium chlorid
e has been tested for simultaneous determination of aerobic and anaerobic b
acteria in the same tube with a soil specimen placed into a meat infusion b
roth. In this experiment, aerobic bacteria form the surface film, and anaer
obic bacteria develop inside the growth medium. The parameters of the exper
iment (meat infusion broth as the growth medium, a constant temperature of
37 degreesC, and the presence of sodium chloride) provide for the predomina
nt development of soil bacilli, for which the presence of sodium chloride i
s an essential ecological factor. It is shown that two biotopes-forest and
plowland (after deforestation(-developing in adjacent sites with similar (s
oddy-podzolic) soils differ significantly by the growth rates of aerobic an
d anaerobic bacilli. In spite of a higher water content in the soil under m
ixed forest, it contains more aerobic zones than the soil of the plowland,
which favors the growth of aerobic bacteria. Statistical methods and calcul
ations based on the Excel and Statgraphics software prove the significance
of the difference between the two soils with respect to the growth rates of
aerobic and anaerobic bacteria.
