Abstract
Objective. The aim of this study was to investigate the effects of enamel m atrix derivative (EMD) on gene expression of collagen alpha1 (I), osteocalc in, prostaglandin G/H synthase 2 (PGHS-2), interleukin-6, and insulin-like growth factor I in primary mouse osteoblasts. Study design. Primary osteoblasts were digested from 6- to 8-day-old mouse calvaria. Cells were divided into 4 groups and cultured for 24, 48, and 72 hours with a serum-free modified eagle medium as negative control, modified eagle medium with 25 mug/mL EMD, modified eagle medium with 100 mug/ml EMD , and modified eagle medium plus 10% fetal bovine serum as positive control . Gene expression was determined by Northern blot and reverse transcription polymerase chain reaction analysis. Result. EMD enhanced collagen I, interleukin-6, and PGHS-2 expression and d id not stimulate the expression of osteocalcin and IGF-I. Conclusion. These results indicate that EMD might regulate certain gene exp ression during periodontal tissue regeneration.
