A novel rnethod for the production of adenoviral vectors on a scale suffici
ent to support most research applications and early phase clinical trials i
s presented. This method utilizes serum-free cell culture medium and a holl
ow fiber cell culture apparatus. Significantly less time and space are requ
ired than in conventional methods, and the resulting adenovirus is collecte
d in a much smaller volume, simplifying the purification steps. The protoco
l described is a reproducible, convenient, biologically safe, and environme
ntally sound method for the production of adenoviral vectors for laboratory
nse and has the potential to scale-up the adenovirus production for clinic
al use.
