The inflammatory cytokine response of peritoneal and splenic gamma delta T-
cells and macrophages to polymicrobial bacterial peritonitis was investigat
ed. The production of intracellular Tumor Necrosis Factor-alpha (TNF) and I
nterleukin-10 (IL-10) was measured using flow cytometry at 6, 24, and 48 hr
s following cecal ligation and puncture or sham laparotomy. TNF-alpha produ
ction was induced in peritoneal - but not splenic - gamma delta T-cells in
response to both sham surgery and peritonitis. gamma delta T-cells demonstr
ated no IL-10 production. Peritoneal and splenic macrophages demonstrated e
arlier TNF and IL-10 responses to CLP than gamma delta T-cells. We conclude
that gamma delta T-cells are local mediators of inflammation.
