An automated trace analytical system for Guanethidine sulfate in sera
was developed using a liquid chromatograph with column-switching and p
ost-column reaction detection systems. Guanethidine sulfate was post-l
abeled with ninhydrin in alkaline solution, and the reaction products
were monitored with a fluorescence detector. Guanethidine sulfate, an
antihypertensive agent, in rat and human sera was analyzed using this
automated analyzer. The detection limit of guanethidine sulfate was 1.
0 ng/mL, and the calibration curve was linear from 3.1 ng/mL to 1000 n
g/mL with relative standard deviation of less than 3%. The recovery fr
om sera was 99.7 +/- 3.9%.
