A practical strategy for solid-phase synthesis of peptide aldehydes has bee
n developed employing a 10,11-dihydroxy-undecanoic acid (DHUA) functionaliz
ed resin. High loading levels are obtained for amino aldehydes as their res
pective acetals. After classical Fmoc SPPS, treatment of the derivatized re
sin with 95% TFA/H2O affords peptide aldehydes in high yield and purity. (C
