An experimental haploid-diploid system suitable for genetic analysis o
f Pleurotus ostreatus using homokaryotic pellets that represent the cl
one offspring of individual spores has been developed. Allele segregat
ion at six out of seven loci did not differ from the expected one. Sig
nificant variation from the expected 1 : 1 segregation was observed at
two phosphohexaisomerase loci (probably linked). The procedure elabor
ated allows one (1) to significantly increase the efficiency of popula
tion genetic analysis in a large group of organisms, basidium fungi; (
2) to use these fungi as a model system in fundamental and applied res
earch.
