Abstract
Replicatively senescent cells of Saccharomyces cerevisiae were obtained usi ng the 'baby machine' method by immobilizing cells on CovaLinks(TM) NH2 pla tes and allowing them to divide while exchanging medium and removing daught er cells. Centrifugation in a Percoll density gradient was employed for fur ther purification of replicatively old yeast cells. Comparison of senescent cells showing more than 20 bud stars with cells from early stationary cult ure demonstrated a significant reduction of total and reduced glutathione a nd decrease of superoxide dismutase activity during replicative aging of ye ast cells.
