Characterization of a peptide antibody toward hGSTA4-4

A polyclonal antibody was developed that can discriminate human alpha class glutathionetransferase 4-4 (hGSTA4-4). This was accomplished by first dete rmining candidate peptide sequences of the hGSTA4 subunit, which exhibited low homology to other human GST isozymes. The chosen peptide sequence (regi on Met108 --> Asn123) of the hGSTA4 subunit was synthesized and used to gen erate an immunogenic response in chickens. The IgY, purified from the resul ting eggs, demonstrated high affinity toward the peptide. The antibody also demonstrated high hGSTA4-4 specificity when tested by Western blot analysi s with several recombinant GST isozymes. (C) 2001 Elsevier Science Ireland Ltd. All rights reserved.
