Momordicoside A has been determined by solid-phase extraction (SPE) on a En
vi Carb cartridge (3 mt, 250 mg) then high-performance liquid chromatograph
y (HPLC) on a Cis column (4.6 mm x 250 mm, 5 mum particle) with acetonitril
e-methanol-50 mM potassium dihydrogen phosphate buffer, 25:20:60 (v/v), as
mobile phase, at a flow rate of 0.8 mi min(-1), and UV detection at 208 nm.
The analytical method was shown to be highly reproducible, with precision
(as RSD) and accuracy las RMD < 10%, both intra-day and inter-day. Absolute
recoveries were > 90%. The method was applied to the determination of momo
rdicoside A in various tissues from different varieties of bitter melon fro
m different producing areas.
