dDNA extraction of thraustochytrids, common marine unicellular organisms, i
s usually accomplished by either the cetyltrimethylammonium bromide (CTAB)
or proteinase K protocols. A novel lysis buffer protocol for thraustochytri
d total DNA extraction is described. The average isolated total DNA is 20 t
o 40 kb, and DNA samples are suitable for a variety of uses including 18S-r
ibosomal UNA polymerase chain reaction, restriction enzyme digestions, and
amplified fragment length polymorphism analyses. The new protocol is also f
aster than the other protocols.
