The notion of dystroglycan as a simple laminin-binding receptor is increasi
ngly being challenged. New roles and new binding partners are continually e
merging. Recent structural advances have provided exciting new insights int
o the precise molecular interactions between dystroglycan and other key com
ponents of the dystroglycan complex. Coupled with an increasing understandi
ng of dystroglycan function at the molecular level, we are finally beginnin
g to probe the complexities of dystroglycan, not only in disease, but in de
velopment, adhesion and signalling.
